Perspective of Small-Molecule AdipoR Agonist for Type 2 Diabetes and Short Life in Obesity

چکیده
Obesity associated with unhealthy diet and lack of exercise is shown to contribute to the onset and/or aggravation of the metabolic syndrome and diabetes, thus placing affected individuals at increased risk of cardiovascular disease and cancer. Plasma adiponectin levels are decreased in obesity, which causes insulin resistance and diabetes. Therefore, we identified adiponectin receptors (AdipoRs) as the therapeutic target. It was suggested that, similarly to caloric restriction and exercise, activation of the AdipoRs may have the potential not only to improve lifestyle-related diseases but to contribute to prolonged the shortened lifespan on a high caloric unhealthy diet. To this end, we have identified "AdipoRon" as an adiponectin receptor agonist. Indeed, AdipoRon ameliorated diabetes associated with obesity as well as to increase exercise endurance, thus prolonging shortened lifespan of obese mice fed on a high fat diet. Additionally, we have recently determined the crystal structures of the human AdipoRs. The seven-transmembrane helices of AdipoRs are structurally distinct from those of G-protein coupled receptors. It is expected that these findings will contribute not only to the elucidation of the AdipoR-related signal transduction but to the development and optimization of AdipoR-targeted therapeutics for obesity-related diseases such as diabetes.

Moderating Role of Alexithymia in Relationship Between Perceived Social Support, Diabetes-Related Quality of Life, and Glycated Hemoglobin in Patients With Type 2 Diabetes
Background: Diabetes is a complex and chronic disease, i.e. associated with patients’ lifestyle and affects their quality of life. The present study examined the role of alexithymia on the relationship between social support, health-related quality of life, and glycated hemoglobin in patients with type 2 diabetes. Methods: This was an analytical cross-sectional study. The study participants c...

OBJECTIVE Type 2 diabetes and obesity are emerging pandemics in the 21st century creating worldwide urgency for the development of novel and safe therapies. We investigated trace amine-associated receptor 1 (TAAR1) as a novel target contributing to the control of glucose homeostasis and body weight. METHODS We investigated the peripheral human tissue distribution of TAAR1 by immunohistochemis...
متن کاملRole of adiponectin in metabolic and cardiovascular disease
Under disease conditions including obesity (insulin resistance) and diabetes, dysregulation of adipokines such as tumor necrosis factor (TNF)-α, leptin, resistin, and adiponectin contribute to the development of metabolic and cardiovascular disease. Unlike other adipokines, adiponectin has been shown to be a therapeutic target for metabolic syndrome and cardiovascular disease. Circulating level...
متن کاملThe Effect of Biochanin A as PPAR γ agonist on LDL Particles Diameter and Type 2 Diabetic Dyslipidemia
Background and Aims: Small dense low-density lipoproteins (sd-LDL) particles are smaller and heavier than typical LDL ones. They can penetrate into the endothelium of coronary arteries more easily because of their small size. Diabetes mellitus is accompanied by dyslipidemia such as increasing concentration of plasma very low density lipoprotein and sd-LDL. Peroxisome proliferator activated rec...

Introduction: The constantly increasing incidence of type 2 diabetes, probably due to obesity and sedentary life, has led to the increased incidence of macro- and microvascular diabetic complications such as nephropathy. Despite the prompt efforts to develop effective treatments for diabetes and slow the progression of its complications, it is still reported as the most common cause of chronic ...
